Absence of lipemia clearing factor lipase in human adipose tissue.

چکیده

Lipoprotein lipase, or lipemia clearing factor lipase (which is more accurate terminology [l]) , is a heparin-activated enzyme which has been found in rat, beef, and pig heart (2). The adipose tissues of rats, rabbits, and especially chickens contain greater quantities of the enzyme (3). This latter observation led to the suggestion that the major site of lipoprotein lipase activity was in the adipose tissue, where it might function in chylomicron dissolution, and also in the lipolysis of depot fat with resultant free fatty acid release. Clearing factor lipase has also been demonstrated in rat plasma after fat feeding (4), and in the plasma of some humans (5, 6) without the prior injection of heparin, indicating a possible intravascular lipolytic role. The evidence supporting this point of view has recently been summarized (7). In view of these facts, the presence of clearing factor lipase activity in human adipose tissue was investigated.

Clearing-factor lipase in adipose tissue. Studies with puromycin and actinomycin.

1. When adipose tissue from starved rats is incubated in a medium containing glucose, insulin, heparin and actinomycin (5mug./ml.) the total clearing-factor lipase activity of the system increases at least tenfold over a period of 9hr. In the absence of actinomycin, enzyme activity also increases, but to a lesser extent and for only about 3hr. Some enzyme activity appears in the incubation medi...

Effect of nutritional status on rat adipose tissue, muscle and post-heparin plasma clearing factor lipase activities: their relationship to triglyceride fatty acid uptake by fat-cells and to plasma insulin concentrations.

1. In rats in a variety of nutritional states, the adipose tissue clearing factor lipase activity is strongly, positively correlated with fat-cell triglyceride fatty acid uptake. 2. In the same animals, muscle clearing factor lipase activity is inversely correlated with the activity of the enzyme in adipose tissue and with the plasma insulin concentration. 3. In starved animals that are given g...

Distinction of Different States of the Enzyme and the Possible Role of the Fat Cell in the Maintenance of Tissue

1. Incubation of intact epididymal adipose tissue from fed rats at 370 in an albumin solution at pH 7-4 in vitro results in a rapid loss of clearing-factor lipase activity until a low activity, stable to prolonged incubation, is attained. The clearing-factor lipase activity of intact tissue from starved rats, which is initially much less than that of tissue from fed rats, is mainly stable to in...

Changes in the lipoprotein lipase (clearing-factor lipase) activity of white adipose tissue during development of the rat.

The lipoprotein lipase (clearing-factor lipase) activity of the white adipose tissue from rats aged between 1 and 145 days was determined. Five adipose-tissue sites (epididymal, uterine, subcutaneous, perirenal and intramuscular) together with serum concentrations of triacylglycerol, cholesterol and glucose were studied. The pattern of enzyme-activity change was remarkably similar in all the si...

Clearing factor, a heparin-activated lipoprotein lipase. I. Isolation and characterization of the enzyme from normal rat heart.

The importance of plasma lipoproteins in normal and abnormal fat metabolism has stimulated interest in the heparin-induced lipemia clearing system first observed by Hahn (2). As a result of the efforts of several independent investigators (3,4), it is now known that,, upon the administrat’ion of heparin to animals of many different species, an enzyme appears in the blood which is able to “clear...
